Despite the clear loss-of-function effects of specific human genetic variations, understanding the impact of many other variants remains a complex undertaking. Our previous report focused on a patient demonstrating a predisposition to leukemia (GATA2 deficiency), carrying a germline variant in GATA2, which inserted nine amino acids within the region between the two zinc fingers (9aa-Ins). Within a genetic rescue system built with Gata2 enhancer-mutant hematopoietic progenitor cells and utilizing genomic technologies, we conducted mechanistic analyses to compare the genome-wide functions of GATA2 and 9aa-Ins.
